In this episode of the Epigenetics Podcast, we caught up with Yael David from Memorial Sloan Kettering Cancer Center in New York to talk about her work on Effects of Non-Enzymatic Covalent Histone Modifications on Chromatin.   The David lab studies on non-enzymatic covalent modifications of Histones, including Histone glycation and citrullination. These modifications recognize metabolites that are produced in the cell and aid as a sensor for chromatin to quickly adapt to cellular changes. These unique modifications do not have a so-called erasing enzyme, which makes them terminal, rendering these sites inaccessible for further modifications such as methylation or acetylation.    A second area of research in the David lab is Histone H1. The lab has developed a new method to purify Histone H1, superior to the commonly used method of acid extraction which leads to degradation of Histone H1. Effects of Non-Enzymatic Covalent Histone Modifications on Chromatin (Yael David)

Assistant professor at Weill Cornell Medical College and head of the Yael David Lab at Memorial Sloan Kettering Cancer Center, Yael David, joins the podcast for a second time. In simple terms,  epigenetic regulation of gene expression explains why--despite every cell in the body containing the same genetic information--some cells carry out unique functions. This gene regulation and expression sheds light on how phenotypic characteristics vary from individual to individual, whether in humans or other species. The research in David’s lab is focused on the modification of histones, which are the proteins responsible for DNA packing in the nucleus. She explains that long-term changes in response to various extracellular signals can lead to changes in DNA histone structure, which can affect cell division, daughter cells, and whole sets of cells in a certain environment. She discusses how colorectal cancer is epigenetically-driven, the quest to determine what causes the epigenetic change responsible for it, and how that epigenetic change drives the development of this type of cancer. Exploring the Relationship Between Epigenetics and Cancer with Yael David, PhD

Yael David utilizes chemistry and biology in her approach to understanding the impact of environmental factors on  epigenetic structures and mechanism and how they in turn affect our DNA.  In this podcast, she explains  Multiple layers of  epigenetic effects on genes from high to low resolution, The different approaches in understanding what determines  epigenetics, including her own stance that environmental factors are key, and How epigenetics in humans relates to cancer, including how damaged chromatin can drive cancer and enzymatic processes can rewrite those damages and protect chromatin.  Yael Davids is lab head and assistant member in the Yael David Lab in the Chemical Biology Program at Sloan Kettering Memorial Cancer Center. She is also an assistant professor of pharmacology at Weill Cornell Medicine.  She explores multiple questions of how  epigenetic regulation of transcription functions alongside histone packaging and DNA and various determinates.  She explains different schools of scientific thought in what effects the  epigenetics of humans along with her own understanding that environmental cues effect and determine epigenetics. She describes how the environmental model explains things such as disease initiation like diabetes, which isn't explained in other models. In this conversation, she discusses how epigenetics change gene function from the changes in DNA itself from protein recruitment to histones, structures that help compact the DNA.
